10 mg/kg/day amodiaquine once a day for 3 days given with 4 mg/kg/day artesunate

Major Interactions

Coadministration with zidovudine is associated with high rates of neutropenia.

Nevirapine-containing ART regimens are associated with reduced amodiaquine exposure.

Efavirenz coadministered with amodiaquine has been associated with hepatotoxicity.

Pharmacology

Antimicrobial class: 4-aminoquinoline

Average serum half life: Approximately 9 days in children with malaria

Metabolism: Rapid first-pass hepatic metabolism to the biologically active metabolite desethylamodiaquine (DeAQ) mediated by cytochrome P450 (CYP2C8).
